Deterministic Lateral Displacement coupled to bio-targeting aptamers for the sorting of Extracellular Vesicles from complex biological samples

Résumé

EVs, biological nanoparticles secreted by cells, appeared more and more attractive as biomarkers accessible by liquid biopsy. However, their isolation method using ultracentrifugation is still not standardized. This paper reports on a microfluidic method for the isolation of Extracellular Vesicles (EVs) based on Deterministic Lateral Displacement (DLD) size sorting coupled to a bio-targeted sorting. This affinity aspect is achieved using specific DNA aptamers directed through EV membrane proteins. EV sorting using DLD device is compared to standard ultracentrifugation isolation in terms of size and concentration. In a second part, aptamer grafting strategy into DLD is demonstrated using CO2 supercritical fluid technology.
